The former Devolution Cabinet Secretary made the announcement during a Jubilee meeting at La Mada Hotel, Nairobi on Monday.
The meeting was to spearhead the merger of all Jubilee parties in Nairobi.
Nominated MP Johnson Sakaja who is also the TNA Chairman and former Starehe MP Margaret Wanjiru also declared their interest in the seat.
"I'm running for Nairobi Governor seat. Kindly treat this as not being a rumour and hopefully my running mate will be from Central Kenya," Sakaja said.
They will battle it out against current Governor Evans Kidero.
Waiguru's declaration comes after she told awomen's gathering at Ufungamano Houseshe would pray and consult before making a decision to enter politics.
The 200 women, drawn from the 17 constituencies in Nairobi, urged Waiguru to join politics and complete the projects she initiated at the National Youth Service.
A week later, the former CS indicated she was consulting President Uhuru Kenyattaover the decision.
